The ABI PRISM® 7700 Real Time PCR System is a fully integrated system for real-time detection of PCR, including a built-in thermal cycler (the venerable ABI 9600), a 10 mW Argon Ion Laser to excite fluorescence, a 64x512 Pixel CCD detector, and real-time sequence detection software with the Apple data system. • This is only one of two laser-excitation Q-PCR systems ever made. Each well is excited by a dedicated fiber, so 96 sample fibers and 3 calibration fibers are used in the instrument. This optical design offers exceptional detection sensitivity and eliminates any cross-talk. The other instrument is the next-generation 7900HT. We expect these instruments to sell at ~$15,000. Quick-Start Manual User Guide Read on: The ABI Prism 7700 Sequence Detection System is a complete, real-time PCR system that detects and quantitates nucleic acid sequences. In real-time PCR, cycle-by-cycle detection of accumulated PCR product is made possible by combining thermal cycling, fluorescence detection, and application-specific software in a single instrument. Multicolor detection provides the flexibility to perform a variety of applications including gene expression analysis, pathogen quantitation, single nucleotide polymorphism (SNP) genotyping, and plus/minus assays that utilize internal positive controls. Real-time PCR measures PCR amplification as it occurs, allowing quantitative measurements to be made in the highly reproducible exponential phase of PCR. The higher the starting copy number of the nucleic acid target, the sooner a significant increase in fluorescence is observed. This revolutionizes the way one approaches PCR-based quantitation of DNA and RNA. Applied Biosystems has developed two types of chemistries used to detect PCR products using Sequence Detection Systems (SDS) instruments: TaqMan chemistry and SYBR Green I dye chemistry. TaqMan probe-based assay chemistry provides outstanding specificity and sensitivity, and the ability to multiplex reactions for real-time quantitation and SNP genotyping assays. The SYBR Green I Dye assay chemistry, while not as specific, provides an economical alternative for target identification. |
